
Enumeration of intestinal
enterococci in 24 hours
ENTEROKAT® is a fast and easy-to-use test system for the detection and enumeration of intestinal enterococci (genus Enterococcus) in water samples. The test is suitable for a wide range of waters, especially drinking water, bathing water and similar with high background bacterial counts that might include an appreciable amount of suspended matter. It allows the detection of enterococci at 1 CFU / 100 ml with definitive results within in 24 ± 2 hours in the presence of heterotrophic bacteria in numbers as high as 1 × 10^6 per 100 ml sample.

Test Principle
ENTEROKAT® utilizes Ortho-Nitrophenyl-ß-D-glucoside as a nutrient indicator and incorporates a specifically designed blue background color in its formulation. When the substrate is metabolized by intestinal enterococci, the sample turns from blue to green to indicate positive detection. After incubation, sample wells (or tubes) that have a green color are considered positive for intestinal enterococci.
As compared to the conventional detection and enumeration of intestinal enterococci by the membrane filtration method according to EN ISO 7899-1 the ENTEROKAT® test system offers multiple advantages:

Application – Instructions for use

Presence / Absence: Add 1 bag of ENTEROKAT® to 100 ml of water sample. Close the sample vessel and shake until the reagent is completely dissolved.

For presence / absence testing incubate the closed vessel for 18-22 hours at 36°C +- 2°C and read-off the result. The vessels should have room temperature before incubation. If turned yellow coliforms are present. If it shows fluorescence under UV-light Escherichia coli is present.

Add 1 bag of ENTEROKAT® to 100 ml of water sample. Close the sample vessel and shake until the reagent dissolves.

For quantitative testing pour the 100 ml sample into the Enumeration Tray.

Seal the Enumeration Tray with the COLIKAT® Enumeration Tray Sealer or another enumeration tray sealing device.

Incubate the sample 18–22 hours at 36 °C ± 2 °C. Count the yellow wells of Enumeration Tray (= coliforms). Count the yellow wells of the Enumeration Tray that show fluorescence under an UV light (365m) in a dark environment (= Escherichia coli).

Transform the count results to the number of CFU in the sample by using the MPN table in the appendix to EN ISO 9308-2.
